This core operates and maintains equipment needed for preparation, quantitation and analysis of DNA and RNA. Equipment is available to support experimental workflows, including DNA sequencing, real-time PCR amplification, and single-cell applications. Each technique has specific applicability depending on the type of samples and the experimental design.

QuantStudio 6 (96-well plates only)
Real-time quantitative PCR system. Can only run 0.2mL semi-skirted plates. This specific instrument is programed to ONLY run the 96-well heating block. Training required.

Qubit Fluorometers detect dyes bound to nucleic acids or other biomolecules. Used for RNA and DNA quantitation and quality in the preparation of sequencing libraries. Training required.
DNA library sequencing instrument. Reagent kits purchased separately by user. Please coordinate with Brad Best (bjbest@versiti.org) for use of instrument, and the Bioinformatics core (bioinformatics@versiti.org) for data export and management. Training required.
The Chromium X controller is a microfluidics-based system for single-cell capture that runs all 10x single-cell assays in standard, low- and high-throughput modes. Compatible with CITE-Seq (Cellular Indexing of Transcriptomes and Epitopes by Sequencing). Reagent kits and processing plates are purchased. Training required.
The BD Rhapsody Single-Cell Analysis System allows capture of 100-100,000 cells on a microwell-based cartridge for multiomics experiments. Compatible with barcoded antibodies for quantitative detection of surface and intracellular antigens by CITE-Seq. Training required.
The ICELL8 platform enables automated dispensing and isolation of single cells in nanowell cartridges for downstream analysis. The instrument allows visual inspection of isolated single cells to distinguish live from dead cells and identify multinucleated cells. Large-bore nozzle suited for isolation of fragile, large cell types. Compatible with SMART-Seq, ATAC-Seq and TCR profiling. Training required.
Highly versatile tissue disruption with single-use gentleMACS tubes. Enables generation of homogenates or single-cell suspensions in a closed, sterile system. Eight samples can be individually processed at a user-defined temperature.
